Abstract: Membrane dynamics of Procyclin, a GPI-anchored protein in Trypanosoma brucei has been studied for the first time by z-scan FCS and FCS Diffusion Laws. The τ0 for procyclin is ~6.8 ms which indicates a cholesterol-dependent nanodomain confinement which on depletion by MβCD gives a τ0 of ~1.6 ms and is ~2.1 ms at 24 hours post CC2D-RNAi and reduces further to ~0.6 ms at 48 hours post CC2D-RNA interference. Dynamics after knockdown of CC2D by RNA interference resembles cholesterol depletion by MβCD on plasma membrane of Trypanosoma brucei.
